    Title: Integrated Analysis of DNA Methylation and Mirna Expression Data To Construct Epigenetic-Regulated Pathways In Non Small Cell Lung Cancer

    Abstract: Experimental studies suggested that there are a role of epigenetic silencing which can lead directly to the formation of cancer. Abberant DNA methylation is one of epigenetic silencing related to downregulation or upregulation miRNA expression in lung cancer cells. However, there is still few comprehensive study in silico study concerning epigenetic role in DNA-methylation and miRNA events.
    We proposed integrated model approach using data mining, statistical spearmen correlation coefficient, meta-analysis computational, cancer target gene database (Mirtar, Tumor Associated Gene (TAG) database and Memorial Sloan-Kettering Cancer Center database, GEO profiles), and cancer disease database (mirtarbase, mir2disease, and HMDDV2) to see epigenetic relationship between DNA Methylation located in their 5,000 bps upstream region of miRNA in Non Small Lung Cancer (NSCLC) using The Cancer Genome Atlas database (TCGA).
    We discovered 81 miRNA query expression was correlated by DNA methylation within 5000 bp upstream. From 81 miRNA we found 33 miRNA might be epigenetic-regulated specific in NSCLC. Sixteen of miRNAs: hsa-let-7g, hsa-mir-145, hsa-miR-200c, hsa-mir-210, hsa-mir-429, hsa-mir-197, hsa-mir-141, hsa-mir-219-1, hsa-mir-212, hsa-mir-34a, hsa-mir-148b, hsa-mir-625, hsa-mir-330, hsa-mir-196a-1, hsa-mir-93, and hsa-mir-135a-1 were confirmed that these miRNA function were involve in the lung cancer. In conclusion, our integrating analysis may help to discover potential direct DNA-methylated and miRNA epigenetic events in lung cancer.
