Color rice had been used as healthy food in traditional Chinese medicinal treatment as people believe purple rice content rich of anthocyanins. Current some research results indicates that anthocyanins have anti-oxidant activity, lower cholesterol, inhibit cell proliferation, promote apoptosis and inhibit cancer cells to rodents and rabbits porridge artery sclerosis and other physical activity. Anthocyanin is unstable in normal environment, especially after cooking the effective component of anti-oxidant material will denatured. The purpose of this project is to develop inexpensive and stable extraction methods for anthocyanin production platforms. The anthocyanin composition, molecular structure, stability and possibility in applied to human health improvement will also be examined. The backcross breeding protocol will be applied for transfer the anthocyanin gene into the current commercialize cultivars. The line which is known rich of anthocyanin content will be use as donor parent and cross to the current high-yield varieties to increase yields potential and stability and also produce high quality anthocyanin. The use of solvent, solvent concentration, acid concentration which will affect anthocyanin extraction and recovery rate will also assay in this project. The stability under the acidic molecular units is required to be discussed further in the project. The overall goals of this project are: 1) generated a purple rice cultivar with good morphology, yield potential, stability and good anthocyanin quality; 2) identified the major component and molecular structure of those anthocyanins extract from this purple rice; 3) study of anti-oxidation mechanisms of purple rice extract; 4) study of mechanisms of purple rice extract on oral cancer cell metastasis.
