    Title: The Alignment and Analysis of Papaya Ringspot Virus Coat Protein Gene Sequence and a Case Study of Transgenic Papaya Cultivation in Nantou County

    Abstract: Papaya ringspot virus which had been found in Hawaii since 1945 is
    the main puthythogene of papaya cultivation. In Taiwan, the papaya
    ringspot virus was found near Kaoshing and Pingtong River sides in 1945 and it was then spread out quickly all over Taiwan papaya cultivation fields. Dr. Shi-Tong Yi transformed the papaya virus coat protein gene PRSV HA5-l , into the cells of papayas in 1987, and then he breeded the first formed anti-PRSV papaya cultivars.
    According our regulation, the trangene papaya are not allowed planted in the farmer's field today. The Agriculture and Food Agency , Council of Agriculture , Executive Yun and the related departments announced that there were no transgenic papayas in the markets. This research regards investigating five Nantou county farmers who plant papayas, and also nine farms which cultivated the papaya without using a nethouse. Through taking samples and exams by peR in the laborato句, it was found that there were six places which farmers planted transgenic papayas. The place near Nan-Gang Junior High School, the Bi-San Road of Caotun, near the Gulf Playground in Zhang-Xing Village, Guoshing Township, Mingjian Township were found to have transgenic papayas. The tatal plottage was nearly 2.1 acres with Mingjian Township had the largest plantation.
    Besides, we compare the gene of PRSV coat protein sequence on NCBI BLASTn for analysis. The result is that over eighty percent of sequences have twenty-two similar nucleotide sequences. After drawing the phylo tree, gene of PRSV coat protein sequence was divided into two groups---the Asia group and the American group. Aligning with the eleven similar sequences of PRSV coat protein sequences , the PRSV coat protein sequences and the Taiwan System---X97251 and X78557 are similar to 99%. The length of PRSV coat protein sequences is organized by l070 oligonucleotides. The length of X9725l sequence of Taiwan YK System is organized by l0326 oligonucleotides. The length of X78557 sequence is organized by l0326 oligonucleotides. Comparing these sequences, the different part is in the third oligonucletide. The PRSV coat protein sequence appears C, but the Taiwan YK System appears T. The other sequences are the same. It means that the PRSV coat protein system and the Taiwan YK System have the very high possibility of the homology.
    Through interviews with farmers and the investigating of the places which grew the papayas without using a net house, it was actually proved由at transgenic papayas plant exist in Taiwan. The result of my research was quite different企om the information企om the Agriculture and Food Agency. It is worthy to be investigated by the related departments. The Agriculture and Food should control企om the resources and furthermore monitor and sample the planted papaya trees of farms all over Taiwan. Through the largest range examination of farms and according to the result of analysis, the government should deal with the problem immediately. Then the consumers' rights can fully be protected.
