    Title: 利用中醫藥資料庫開發中風治療新藥

    Abstract: 中風是無法有效預防的嚴重疾病,而N-Methyl-D-aspartate receptor(NMDAR)所釋放的興奮性毒性為中風的首要致病原因。近期中風有突破性的發展,NR2A有著神經保護功能;相反的,NR2B有著誘導神經死亡的功能。本實驗希望挑選出一種可以同時促效NR2A以及抑制NR2B的天然化合物,利用中醫藥資料庫跟NMDAR作進行虛擬藥物篩選,挑出Dencicihine(三七素)、Alliin(蒜胺酸)與對照分子比較在NR2A與NR2B上有高度相似的鍵結。分子動態模擬結果發現三七素與蒜胺酸在接上NR2A不僅有較高的氫鍵佔有率,更有側邊陽離子作用力和鹽橋形成,帶來更高的穩定性。根據我們的研究結果,中藥分子三七素與蒜胺酸能同時對NR2A與NR2B產生作用,其雙重目標特性,將有助於雙重目標藥物的開發,使治療更為專一,降低不適用藥物所帶來的負面併發症風險。
