近年來,台灣食安問題層出不窮,進而促進社會大眾食安意識提高。沙門氏菌 (Salmonella) 及單核細胞增生李斯特菌 (Listeria monocytogenes) 為食品重要的病原菌,在許多即食食品、加工肉類、奶酪、乳製品和雞蛋中皆可發現,為了防止食用受汙染之食品危害健康,因此,開發靈敏、快速的檢測方法至關重要。 本研究利用生物傳感器,結合電化學阻抗圖譜 (Electrochemical impedance spectroscopy, EIS),在牛奶中檢測沙門氏菌及單核細胞增生李斯特菌,可利用即時、快速、靈敏、操作簡易等優點,進行有效的檢測方法。結果顯示,金電極晶片結合EIS技術,利用抗原抗體的特異性,可在短時間內得到可靠的結果。將方法優化後,應用於檢測牛奶樣品中Listeria monocytogenes及Salmonella spp.,其結果靈敏度為N×100 CFU/mL。 In recent years, Taiwan food safety problems have emerged one after another, which in turn has promoted the improvement of public food safety awareness. Salmonella and Listeria monocytogenes are important food pathogens, which can be found in many ready-to-eat foods, processed meat, cheese, dairy products and eggs. In order to prevent the consumption of contaminated food from endangering health , therefore, it is crucial to develop sensitive and rapid detection methods.In this study, a biosensor was used in combination with Electrochemical impedance spectroscopy (EIS) to detect Salmonella and Listeria monocytogenes in milk. The advantages of instant, rapid, sensitive, and easy operation can be used for effective detection method.The results show that the gold electrode chip combined with EIS technology can obtain reliable results in a short time by utilizing the specificity of antigen and antibody. After the method was optimized, it was applied to the detection of Listeria monocytogenes and Salmonella spp. in milk samples, and the sensitivity was N�100 CFU/mL.
