菇類已在台灣發展超過百餘年,現今在各大賣場皆可看到各式各樣菇類身影於貨架上,可說是國民不可或缺的營養來源之一,並且關於菇類研究也日益趨增,目前已許多關於提高產量以及縮短栽培時程的研究,大部分的方向皆為更改基質的配方或是使用不同的接種方式。本研究之目的為: (1)測試出合適的開袋方式; (2)找出秀珍菇合適的萃取條件; (3)對秀珍菇萃取物進行抗氧化實驗; (4)測試萃取物是否對於黑色素癌細胞具有毒殺性。研究結果顯示,最佳的開袋方式為開兩條線的方式,冷水最佳萃取固液比為1:40,而熱水為1:50,抗氧化物質含量熱水萃取較高,抗氧化能力冷水較佳,細胞實驗結果顯示冷水萃取以及熱水萃取皆對小鼠纖維母細胞以及小鼠黑色素瘤皆無毒殺性,在光老化實驗中,得到秀珍菇冷熱水萃取物對於受到UVB損傷的NIH3T3細胞具有修復的作用。此研究證實了秀珍菇較佳的開袋方式,未來可繼續往高產量縮短栽培時程繼續發展,萃取物對於正常細胞不具有毒殺性,作為保健食品可增加營養為具有潛力的商品原料。 Mushrooms have been developed in Taiwan for more than a hundred years. Nowadays, kinds of mushrooms can be seen on the shelves in supermarkets. Mushroom is one of the indispensable sources of nutrition for people, and research on mushrooms is also increasing. Pleurotus sajor-caju has high economic value. There have been many studies on increasing yield and shortening the cultivation time. Most of the research directions are to change the formula of the substrates or to use different inoculation methods. The purposes of this thesis are: (1) To study suitable ways of bag opening for Pleurotus sajor-caju; (2) To compare the extraction methods of Pleurotus sajor-caju; (3) To analyze antioxidant contents and abilities of Pleurotus sajor-caju extracts; (4) To evaluate the toxicity of Pleurotus sajor-caju extracts to cancer cells. The experimental results showed that the best way to open the bag is two-line opening. The best extraction concentration of cold water is 1:40 and hot water is 1:50. The antioxidant content of hot water extract was higher than cold water extract of Pleurotus sajor-caju, and the antioxidant capacity of cold water extract is higher than hot water extract. The results of cell experiments showed that both cold water extract and hot water extract are nontoxic to cells. This experiment confirmed the better way to open the bag of Pleurotus sajor-caju, In the future, we can continue to develop high yields and shorten the cultivation time. The extracts of Pleurotus sajor-caju was not toxic to normal cells, it is a potential commercial raw material health food for people. In the photoaging experiment, the cold and hot water extract of Pleurotus sajor-caju mushroom has a repairing effect on NIH3T3 cells damaged by UVB. This research confirms that the better way of opening the bag of Pleurotus sajor-caju mushroom can continue to develop in the future with high yield and shorten the cultivation time. The extract is not toxic to normal cells, and could be as a healthy food, it can increase nutrition and become a potential commercial raw material.
