    Abstract: 本研究的目的是探討百里酚對順鉑所誘導的腎毒性的治療活性。百里酚是一種天然的易揮發性單?酚類化合物,從食品到農業化學品皆有廣泛應用,許多研究表明百里酚具有抗微生物、抗氧化、抗發炎等藥理活性,可能對人類健康產生有益的影響。本實驗設計為連續14天分別給予小鼠低劑量50 mg/kg與高劑量100 mg/kg的百里酚,在第14天於小鼠腹膜內注射15 mg/kg的順鉑,於72小時後犧牲小鼠進行觀察。實驗結果發現百里酚改善由順鉑引起的血清肌酸酐和血液尿素氮的增加。透過西方墨點法測定轉錄因子Nrf2、血基質氧化?-1及發炎小體蛋白,百里酚的給予增加了抗氧化蛋白的表現並降低了由順鉑引起的發炎小體蛋白及發炎細胞激素表現,顯示對氧化壓力及發炎反應具有抑制作用。這些發現表明百里酚可能是一種有效的腎保護劑,能減少順鉑所誘導的急性腎毒性。
    The aim of this study was to explore the protective effect of thymol on acute kidney injury induced by cisplatin. Thymol is a natural volatile monoterpenoid phenol compounds, which is widely used from food to agrochemicals. Many studies shows that thymol has antimicrobial, antioxidant, anti-inflammatory and other pharmacological activities, and may have beneficial effects on human health. Mice were intraperitoneally administered cisplatin at a dose of 15 mg/kg to induce acute kidney injury and orally administered thymol at doses of 50 and 100 mg/kg for 14 days. Seventy two hours later, mice were sacrificed to make observations. The results of the experiment found that treatment with thymol significantly ameliorated the increase in serum creatinine and blood urea nitrogen (BUN) induced by cisplatin, confirming that thymol has the effect of decreasing kidney damage. The levels of Nuclear factor erythroid 2-related factor 2 (Nrf2)、Heme oxygenase-1 (HO-1) and NLR family, pyrin domain containing protein 3 (NLRP3) protein were determined by Western blot, shows that oral administration of thymol protected against the cisplatin-induced injury, significantly increase the expression of antioxidant protein and reduced the expression of NLRP3 and also inflammatory cytokines, indicating the inhibition of oxidative stress and inflammation. These results suggest that thymol is an effective nephroprotective agent that may be used to ameliorate cisplatin-induced nephrotoxicity.
