    Title: 松杉靈芝萃取物對口腔癌細胞轉移的抑制作用

    Abstract: 口腔癌是台灣常見的惡性腫瘤之一,在台灣十大癌症中排行第五名。由於口腔癌容易轉移及復發,造成治癒率低,尚缺乏有效的治療方法。近來由於中藥全球化聯盟(Consortium for Globalization of Chinese Medicine; CGCM)的全球推廣,加上中藥本身的低毒性及長久以來的人體試驗,使得中藥成為現今輔助治療疾病的首選。靈芝是中草藥中最廣為人知之一,並有抗腫瘤、抗發炎、抗老化等等的功效。而在許多研究顯示,靈芝可以抑制多種癌症的轉移,但在口腔癌的轉移上尚不清楚。因此,本研究的目的是探討靈芝抗口腔癌轉移的效果。本實驗室先前研究的結果顯示靈芝可以藉由細胞凋亡以及自噬作用來抑制口腔癌細胞(HSC-3)的生長作用。而在本研究的細胞遷移試驗(Wound healing & Transwell assay)中發現靈芝能夠抑制口腔癌細胞的移動性,另外,在侵襲試驗(Invasion assay)裡,發現靈芝可以降低口腔癌細胞的侵襲能力,而在Zymoraphy實驗中發現靈芝能夠降低口腔癌細胞中MMP-9的活性,也就是說,靈芝可以抑制口腔癌細胞分解細胞外基質的能力。這些結果顯示靈芝有著影響口腔癌細胞轉移的潛在能力,且未來在治療口腔癌中可成為有效的輔助治療藥物。
    Oral cancer is one of the common malignancies in Taiwan, ranking the fifth in Taiwan's top ten leading cancers. There is no effective treatment for oral cancer due to the ease of metastasis and recurrence, resulting in a low cure rate. Recently, due to the global promotion of the Consortium for Globalization of Chinese Medicine (CGCM), coupled with its low toxicity and long-term human trials, traditional Chinese medicine has become the first choice for the adjuvant treatment of diseases. Ganoderma is one of the most well-known herbal medicines with anti-tumor, anti-inflammatory, anti-aging and other effects. Many studies have shown that Ganoderma can inhibit the metastasis of many types of cancers, but it is still unclear about its effect on the metastasis of oral cancer. Therefore, the purpose of this study is to investigate the effect of the ethanol extract of Ganoderma tsugae (GTEE), a well-known Ganoderma cultivated in Taoyuan, Taiwan, on the metastasis of oral cancer. Previous studies in our laboratory showed that GTEE could inhibit the oral cancer HSC-3 cell proliferation through induction of apoptosis and autophagy. The Wound healing & Transwell assays demonstrated that GTEE could inhibit the motility of oral cancer cells. Furthermore, the invasion assay elucidated that GTEE could also inhibit the invasion of oral cancer cells. In addition, GTEE was found in Zymoraphy experiments to decrease the MMP-9 activity in oral cancer cells, showing that GTEE could suppress the ability of oral cancer cells to degrade extracellular matrix. These results indicate that GTEE has the potential to affect the metastasis of oral cancer cells, and may provide an effective adjuvant therapeutic strategy in the clinical treatment of oral cancer.
