    Title: The Protocol for Dendrobium Officinale Mass Propagation

    Abstract: There are more than 1,600 species of Dendrobium in the world. Some of them had been reported can be used as medicine herb, for example D. officinale, D. huoshanense, D. moniliforme and D. nobile. Dendrobine is the alkaloids extract from Dendrobium spp. which can cure analgesic, antipyretic, reduce heart rate, high blood pressure and slow breathing. A ccording to previous research, D.officinale can increase NO and TNF-α increased macrophage secretion and phagocytosis to enhance the immune mechanism, it can also promote islet cells make insulin and glucagon, to achieve the effect of lowering blood pressure. Heavily harvested due to its medicinal effects, D. officinale is rarely can be seen in nature also markets appeared to be short in supply, and it also found out cannot year-round planting in Taiwan. The cultivation environment such as temperature tolerance of D. officinale is from 18℃ to 30℃, and it will stop growing at temperature more than 35℃. Therefore, the method to cultivate large amount of D. officinale is an important topics at present.
      The artificial reproduction of D. officinale was carried out by tissue culture and environment. The environment result shows D. officinale grow 0.64 cm/month in red + white light (3474 lux), the highest compare with other light intensities. A capsule of D. officinale germination can sprout 1713 seedling. In addition, the buds grow optimally in red light (1304 lux) which is 1 unit/24 days. the seeds are from capsule of D. officinale on MS agar for germination, than inoculation to seedling medium until adult. But the genetic will be changed by this method. In this research, we used high temperature treatment to induce D. officinale stem adventitious buds. The result shows the probability of inducing adventitious buds was 50.28%. The growth regulators also effect the growth of the D. officinale buds. The result showed 2ppm 2, 4-D + 0.5ppm 6-BA MS medium have the fastest growing, the growth rate is 0.026 cm/days. Using ½ MS agar without growth regulators is the most effective way to induce lateral buds, an average of 6 days can induce a bud. Combining all the data, it is possible to induce the buds for D. officinale in high temperature and propagation it in medium low temperature such as 25℃ in ½ MS agar without growth regulators.
    Combining all the data, 60,000 D. officinale can growth 2,239,470 in first year, then get 39,188,666 plants in second years. Through it is possible to cultivate the D. officinale in plant factory and have the economic benefits of the market.
